It would be wise to start with a smaller, more dedicated group of Tweeters and build your follow group there.

Try a simple post, a note, or an electronic mail to your contacts and friends stating that you have joined Twitter and you’d like to follow them, and suggest for them to follow you back. Usually follows work both ways and the prospective follower would enjoy a follow as well. This will help build online relationships and repertoire. It will not hurt to invite new users to Twitter either. Tell them a little about it and allow them decide what is fitting for them.
You are more likely to get followers from your friends' and coworkers' followers than you are from random strangers. Here is why Twitter is so social - when there are retweets or replies to your update, it shows up on another feed, which in turn shows on the screens of whoever follows that feed. To give you some comparisons, if you tweet something and have 20 followers, and just one of your followers retweet it, that is now showing up to your 20 followers, as well as however many followers the person had that retweeted. You can see that depending on the number of followers they have, this has the potential to reach thousands of people. Social marketing in it’s finest.
However, getting the followers is only half the challenge. In order to keep your followers active and engaged, you'll need to post exciting content. Post what you love, but make sure that you include something you think others will enjoy. The tweets have to be easy to read and understand. Never post boring updates. If you wouldn't want to read it on someone else's Twitter, why would someone want to read it on yours? Hearing about the big lunch you just ate doesn't appeal to anyone. Good luck and happy Tweeting!
